In Vitro Effect of TNF- and IFN- in Retinal Cell Infection with Toxoplasma gondii

چکیده
METHODS. Rat retinal cells, retinal pigment epithelial (RPE) cells, and retinal Müller glial (RMG) cells were in vitro infected with T. gondii RH strain tachyzoites. Cultured cells were stimulated with various concentrations of TNFand IFN. The effect of TNFand IFNin T. gondii invasion and replication between retinal cells was determined through two different methods: measuring [H]-uracil incorporation and counting infected cells by microscopic examination.

A serological and parasitological study of Toxoplasma gondii infection in stray cats of Mashhad, Khorasan Razavi province, Iran
The aim of the present study was to determine seroprevalence of Toxoplasma gondii infectionin stray cats and correlation with oocyst shedding and IFN-γ concentration. From April to August 2016, one hundred fifty-nine stray cats were captured from various localities in Mashhad area. The blood and fecal samples were collected from each cat. The serum samples were examined to detect antib...
متن کاملProtective Efficacy of the Killed Toxoplasma gondii Vaccine in Nano-alum Adjuvant
Background and purpose: Toxoplasma gondii (T. gondii) as a causative agent of toxoplasmosis, is an obligate intracellular parasite which is excreted by the cat's feces. Efforts have been made for the development of toxoplasmosis vaccine, but none led to developing a vaccine with protective immune response to the parasite. Adjuvants are essential for vaccine formulation to create strong immune r...
متن کاملCD40 restrains in vivo growth of Toxoplasma gondii independently of gamma interferon.
CD40-CD154 interaction is pivotal for resistance against numerous pathogens. However, it is not known if this pathway can also enhance in vivo resistance in gamma interferon (IFN-gamma)-deficient hosts. This is an important question because patients and mice with defects in type 1 cytokine response can control a variety of pathogens. While blockade of endogenous CD154 resulted in a remarkable i...
متن کاملIn vitro investigation of host resistance to Toxoplasma gondii infection in microglia of BALB/c and CBA/Ca mice.
Toxoplasmic encephalitis (TE) is a life-threatening disease of immunocompromised individuals and has increased in prevalence as a consequence of AIDS. TE has been modeled in inbred mice, with CBA/Ca mice being susceptible and BALB/c mice resistant to the development of TE. To better understand the innate mechanisms in the brain that play a role in resistance to TE, nitric oxide (NO)-dependent a...
متن کاملSingle nucleotide polymorphisms in the interferon gamma gene are associated with distinct types of retinochoroidal scar lesions presumably caused by Toxoplasma gondii infection
The association of single nucleotide polymorphisms (SNPs) in the interferon (IFN)-γ gene ( IFNG ) with different types of retinal scar lesions presumably caused by toxoplasmosis were investigated in a cross-sectional population-based genetic study. Ten SNPs were investigated and after Bonferroni correction, only the associations between SNPs rs2069718 and rs3181035 with retinal/retinochoroidal ...
